چکیده
Background: Estradiol plays a pivotal role in establishing endometrial receptivity by regulating genes involved in angiogenesis, including vascular endothelial growth factor (VEGF) (1-3). However, the relationship between circulating estradiol concentrations and endometrial VEGF protein expression during the pre-implantation period remains unclear. This study investigated the correlation between serum estradiol concentration and endometrial VEGF protein expression in ovarian-stimulated mice immediately before implantation. Methods: Twenty adult female mice were assigned to two groups: 1) control and 2) ovarian stimulation (7.5 IU human menopausal gonadotropin (HMG), followed 48 hours later by 7.5 IU human chorionic gonadotropin (hCG)) (4, 5). Pearson's correlation analysis was performed to evaluate the association between Serum estradiol concentrations, quantified by ELISA, and endometrial VEGF protein expression, determined by immunohistochemistry and Western blot analyses. Findings: Significant positive correlations were identified between serum estradiol levels and VEGF protein levels measured by immunohistochemistry (IHC) and Western blotting. In the control group, serum estradiol levels were positively correlated with VEGF protein expression assessed by IHC (r = 0.637, P = 0.021) and Western blotting (r = 0.551, P = 0.021). Stronger positive correlations were observed in the ovarian-stimulated group. Conclusion: Serum estradiol levels were positively associated with endometrial VEGF protein expression in ovarian-stimulated mice immediately before implantation, supporting a potential role for estradiol in the regulation of pre-implantation endometrial angiogenesis.
