چکیده :
Background: Angiogenesis-related microRNAs regulate the expression of genes responsible for blood vessel formation, including Vascular Endothelial Growth Factor (VEGF). Angiogenesis is indispensable during pregnancy, as it supports development of a receptive endometrial environment for embryo implantation. Progesterone plays a role in regulating endometrial transformation and promoting angiogenic processes during this period (1-3). This study aimed to examine the relationship between miR-16-5p expression and VEGF levels in the pre-implantation endometrium of mice receiving exogenous progesterone. Methods: Twenty adult female mice were divided into two groups: 1) control and 2) progesterone (1 mg/mouse of progesterone intraperitoneally for 3 consecutive days, 72 hours after day 0) (4, 5). Pearson's correlation analysis was performed to evaluate the association between endometrial miR-16-5p expression, quantified by real-time PCR, and VEGF protein levels, determined by immunohistochemistry (IHC) and Western blot analysis. Findings: Regarding IHC data, miR-16-5p showed a strong positive correlation with VEGF protein expression (r = 0.74, p<0.05 in the control group; r = 0.78, p<0.01 in the progesterone-treated group). Similarly, Western blot analysis confirmed a significant positive correlation between miR-16-5p expression and VEGF protein levels. Conclusion: miR-16-5p may play a role in regulating endometrial angiogenesis through its association with VEGF during the pre-implantation period in the presence of exogenous progesterone.
