چکیده :
Background: Fibroblasts are essential for cutaneous wound healing, with distinct subpopulations contributing to different repair phases. Emerging evidence indicates that neuropeptides, neurotransmitters, and neuroimmune interactions regulate fibroblast heterogeneity and function. This review examines the role of neurogenic regulation in fibroblast heterogeneity during wound healing and its implications for regenerative medicine (1-3). Methods: This narrative review was conducted through a comprehensive search of the PubMed, Scopus, and Web of Science databases for studies published up to 2025. The search strategy included the keywords fibroblast heterogeneity, cutaneous wound healing, neuropeptides, and neuroimmune signaling. Studies investigating the neurogenic regulation of fibroblast subpopulations and the underlying molecular mechanisms were selected, critically evaluated, and synthesized (1-5). Findings: The reviewed studies demonstrated that neuropeptides, including substance P, calcitonin gene-related peptide (CGRP), norepinephrine, and acetylcholine, regulate fibroblast proliferation, migration, extracellular matrix synthesis, and differentiation into myofibroblasts, thereby shaping the functional heterogeneity of these cells (2, 4). In addition, single-cell RNA sequencing (scRNA-seq) studies have identified distinct fibroblast subpopulations with differential responses to neural signals. These subpopulations exhibit specialized roles throughout the wound-healing process, from inflammatory regulation to tissue remodeling and regeneration (1, 3, 5). Conclusion: Neurogenic regulation is a key determinant of fibroblast heterogeneity during cutaneous wound healing. A deeper understanding of neuron-fibroblast signaling pathways may facilitate the development of targeted therapeutic strategies to reduce scar formation, control fibrosis, and advance regenerative medicine.
