چکیده :
Neuroinflammation is a key component of the secondary injury cascade after traumatic spinal cord injury (SCI), yet its dynamic and time-dependent nature complicates anti-inflammatory treatment. Conventional drug administration is further limited by inadequate exposure at the lesion site, rapid clearance, restricted tissue penetration, and unfavorable release kinetics. Nanoparticle-based delivery systems have therefore emerged as a potential strategy to improve local drug availability and provide greater control over therapeutic release within the injured spinal cord. This mini-review examines preclinical studies of nanoparticle-mediated delivery of anti-inflammatory agents in SCI, focusing on corticosteroids, repurposed drugs, and natural compounds such as curcumin. Across experimental models, several nanocarrier-based formulations have been associated with reduced inflammatory activity, improved tissue preservation, and better locomotor recovery. However, interpretation of these findings is complicated by substantial variation in injury models, routes and timing of administration, formulation design, and the choice of comparator groups. Importantly, an improved outcome following treatment with a nanoformulation does not, by itself, demonstrate an advantage over the corresponding free drug. Further studies should include appropriately matched free-drug controls, detailed characterization of drug distribution and release behavior, long-term safety assessment, and validation in clinically relevant SCI models. The clinical potential of nanocarrier-based anti-inflammatory therapy will ultimately depend on whether these systems can provide reproducible improvements in efficacy or safety compared with conventional drug formulations.
