چکیده :
Background: Reconstruction of craniofacial bone defects requires scaffolds that, while providing adequate mechanical strength and appropriate porous architecture, offer a favorable environment for cell growth and the ability to deliver regenerative agents (1-3). In this study, a multifunctional scaffold was designed and evaluated by combining a three-dimensional (3D)-printed polylactic acid/hardystonite-graphene oxide (PLA/HTGO) structure with a simvastatin-containing microporous phase. Methods: PLA/HTGO scaffolds were fabricated using 3D printing based on the optimized formulation (4). To create a microporous structure and enable drug loading, a pectin-quaternized chitosan polyelectrolyte solution containing simvastatin was introduced into the scaffold channels and stabilized by freeze-drying. Structural characteristics and drug release, cellular behavior using MG-63 cells, and bone defect repair capacity were evaluated in a rat calvarial defect model (5). Findings: The incorporation of GO optimized the mechanical and biological properties, while the formation of the polyelectrolyte phase generated a network of micropores within the architecture of the printed scaffold. The simvastatin-containing scaffold enhanced MG-63 cell adhesion, proliferation, and spreading. In addition, in vivo evaluation after 8 weeks demonstrated extensive bone defect regeneration and new bone formation within the defect area. Conclusion: Combining the macroporous architecture provided by 3D-printed PLA/HTGO with a simvastatin-containing microporous pectin-quaternized chitosan phase resulted in a scaffold with complementary structural and therapeutic functions. This system demonstrated suitable potential for application in craniofacial bone regeneration and may provide a basis for further preclinical studies.
