چکیده :
Background: Peripheral nerve injuries often result in significant sensorimotor deficits. While autologous nerve grafting is the current gold standard, its clinical application is hindered by limited donor availability and donor site morbidity [1,2]. Consequently, novel strategies incorporating biomaterial scaffolds, neurotrophic factors, and cell therapies are being investigated as promising alternatives to overcome these limitations and enhance functional nerve regeneration [3,4]. Method: This review aimed to evaluate recent approaches to peripheral nerve regeneration. The roles of natural and synthetic nerve scaffolds, stem cells, Schwann cells, growth factors, and pharmacological agents in nerve repair were examined [3-5]. Finding: Natural scaffolds such as collagen, chitosan, and alginate, as well as synthetic polymers including PLLA, PLGA, and PCL, provide physical support and a favorable microenvironment for axonal growth and guidance [3,5]. In addition, mesenchymal stem cells, adipose-derived stem cells, bone marrow-derived stem cells, and Schwann cells contribute to nerve regeneration by secreting neurotrophic factors, enhancing cell survival, promoting remyelination, and supporting axonal extension [4,5]. Pharmacological agents such as dexamethasone, methylprednisolone, melatonin, acetyl-L-carnitine, vitamin B12, and citicoline have also demonstrated neuroprotective and regenerative effects in experimental studies [4]. Moreover, combination therapies involving growth factors, stem cells, and pharmacological agents appear to produce superior regenerative outcomes [5]. Conclusion: The integration of tissue engineering, cell-based therapy, and pharmacological treatment offers a promising strategy for peripheral nerve regeneration [3-5]. Although further clinical studies are required, these approaches have considerable potential to improve the treatment outcomes of peripheral nerve injuries [2,5].
